Alomone Labs has developed three innovative product lines for studying protein-protein interaction and/or protein co-localization:
Primary antibodies conjugated to biotin or ATTO-fluorescent dyes: These antibodies can be used in immunohistochemistry (IH) and immunocytochemistry (IC) with same species antibodies.
Antibodies raised in guinea pig: These second species antibodies can be used with any other non-guinea pig second species antibodies in immuno-colocalization studies such as IH and IC.
Toxins conjugated to ATTO-fluorescent dyes: These labeled toxins can be used with any antibody to obtain highly specific channel localization.

Immunohistochemical staining of rat frozen brain sections: Guinea pig Anti Kir4.1 Antibody (#AGP-012), (1:300). K ir4.1 staining (green) is detected as clusters in the molecular layer.
|
|
Immunohistochemical staining of rat spinal cord frozen sections: Anti-Connexin-36-ATTO-488 Antibody (#ACC-209-AG). Connexin-36 staining (green) is scattered along layer I of the dorsal horn.

Immuno-colocalization of Synapsin-2 and GABA(A) α1 Receptor in rat striatum.
Immunohistochemical staining of immersion-fixed, free floating rat brain frozen sections using rabbit Anti-Synapsin-2 Antibody (#ANR-015), (1:300) and Guinea pig Anti-GABA(A) α1 Receptor (extracellular) Antibody (#AGP-083), (1:600). Synapsin-2 staining (red) appears in Islands of Calleja. GABA(A) α1 Receptor (green) is also detected in Islands of Calleja and in adjacent neurons. Merged staining demonstrates colocalization in Islands of Calleja while adjacent GABA(A) α1 positive neurons do not express Synapsin-2. Cell nuclei are stained with DAPI (blue).

A Super Potent Blocker of KV1.2 Channel
|
Urotoxin (#STU-200) is a peptide toxin originally isolated from Urodacus yaschenkoi Australian Scorpion venom. Urotoxin is an extremely potent blocker of human voltage-gated potassium channel K V1.2 channels with IC 50 values of 160 pM. However, it also displays activity towards K V1.1, K V1.3 and K Ca3.1 with IC 50values of 253 nM, 91 nM and 70 nM respectively.

LTH Increases KCa1.1 Activity but not Expression
Long term hypoxia (LTH) was found to increase Ca2+ affinity of BK Channels in cerebral artery smooth muscle of both adult and fetal ovine.
Electrophysiological experiments indeed showed that BK channels are significantly more active under LTH. However, immunohistochemical staining of BK channels in ovine arteriole myocytes using Anti-KCa1.1 (BKCa) (extracellular) Antibody (#APC-151) shows that the α subunit is equally expressed among the different groups.

Expression of KCa1.1 in adult and fetal ovine arteriole myocytes.
Immunohistochemical staining of adult and fetal ovine arteriole myocytes using Anti-KCa1.1 (BKCa) (extracellular) Antibody (#APC-151). K Ca1.1 staining (green) is detected similarly whether under normal conditions (right panels) or under hypoxia (left panels).
